Affinity chromatography is a key research tool for the purification of known ligands, including antigens and proteins, and, as an extension, identification of novel interacting partners. Numerous methods exist for the generation of affinity columns, using various ligands and coupling chemistries. One important tool is the covalent coupling of glycoproteins to reactive hydrazides immobilized on a solid support (agarose beads).

This technique is routinely used for the immobilization of monoclonal and polyclonal antibodies with glycosylated Fc domains, as the resulting immobilized antibodies have unobstructed antigen-binding domains.
